When Richard Greico was asked that at the club he snapped back with a no.
Was that just because in the movie he was suppose to be annoyed with being there, or is there a deeper issue with him and Depp?

I always get a chuckle out of that scene. My guess is, Greico thought it was a stupid thing to ask since he had nothing to do with Depp apart from starring in 21 Jump Street. Or, his reaction was meant to show his supposed jealousy (in the movie) of Depp's success post-21 Jump Street.
