After Anthony and Lesly were finished, there was something said about Anthony not being a virgin and since he appears to never leave the house, could it be that he had an affair with Jackie also?

No, I believe Jackie was only doing it with Marty. We can see that because Anthony's relationship with Jackie is not the same as her relationship with Marty.
